主要研究背景及目的

To reduce the residual recurrent risk of acute ischaemic stroke remains a challenge. Inflammation has been associated with the incidence and recurrence of stroke. The recent COLCOT trial demonstrated that low-dose colchicine as an anti-inflammatory agent reduced the risk of cardiovascular events in patients with acute coronary artery diseases. A subsequent meta-analysis showed that the risk of stroke was almost halved in colchicine-treated patients with coronary artery disease. The risk of subsequent stroke within 3 months after the index event accounts for approximately 70% of subsequent strokes within 1 year and 40% within 5 years. However, there is still an absence of evidence regarding efficacy and safety of acute use of colchicine for the prevention of early recurrent stroke.

方法

All efficacy and safety analyses were by intention-to-treat.We compared the difference in event risk between the two trial groups by a shared frailty model, which incorporate the random effect of the trial center on the baseline hazard of Cox proportional-hazards model; hazard ratios and 95% confidence intervals were reported.
